A serpentine draft is a fantasy football term that describes a draft that goes in the opposite order each round. It is the same as a snake draft, which is more popular term.
In a serpentine draft the order of teams drafting flips every round. So the team with the last pick in the first round has the first pick in the second round and so on. The "serpentine" part comes from the shape of the draft and how it resembles the shape of a snake with its wave shape. The serpentine draft allows teams to draft more equally than if each team stayed in the same position each round, which is what the NFL draft does.
